The paper reports the estab li shmcill or mycorrhi za l inrecti on or a non-mycorrhi za l Ri-T-DN A trans rormcd carrot root when co-c ultured with a su rrace sterili zcd swcd potato root scgment coloni zcd by arbusc ular mycorrhi zal (A M) rungus C. ill/rami/ices on minimal M medi um. Extensivc rungal hypha I cmergence rrom eac h cut end of the mycorrhi zal swcet potato root piece was observed in one weck old cullUres. Thes\: hyphae causcd infecti on on contacting the transrormed-carrotroot segment and produced many hyphae and spores both inside and outside the zone of the root aft er 6 week or growth. Axeni call y produced rungal propagules prolirerated on the surface of fresh minimal M medium when sub-cu ltured without any root seg ment. On repeated sub-culturing, thesc propagules did not lose their ability to grow and produced man y juveni le small spore-likc ves icles durin g thc non-sy mbiotic phase. Although these spores were morphologicall y and anatomical ly si milar to their soi l borne counter part s, they were much smaller. When pl aced in the vieinity or a fresh hairy root on the minimal medium or a Sudan grass seedling in sand eu llllre, thc axenica ll y produced AM runga l propagu les caused root in rec ti on, but the inrection characteri stics we re signiri cantl y dirfe rent to the ori ginal culture in terills o r shape (spheri cal vs oval) anc! sizc (20 ~lIn vs 45 pm) or thc intraradical ves icles. and absence or 'H' branches. Sudan grass seedlings inocu lated with the axcni call y cultured rungus showed signiri cantly (P <: 0.05) higher dry weights plant i . When compared to the plants inocu lated wi th sand cultures. the growt h paranlcters and thc pcrcentage inrec ti on werc not signifi cant ly dilTerent. However, when bot h sources o r inoc l'la werc uscd together, a sy nergistic cl'fcct on plant growth as we ll as root infection was obscrvcd.

In vitro interaction studies between Glomus intraradices and Armillaria mellea in vines

An interaction study was performed with mycorrhizal plants of the grapevine rootstock Richter 110 (Vitis berlandieri Planch × Vitis rupestris Scheele) and the root pathogenic fungus Armillaria mellea (Vahl:Fr.) P. Kumm using an autotrophic in vitro culture system. Micropropagated plantlets were transferred to Petri plates with MSR medium lacking sugar and vitamins. Inocula of Glomus intraradice...

Heavy-metal stress and developmental patterns of arbuscular mycorrhizal fungi.

The rate of global deposition of Cd, Pb, and Zn has decreased over the past few decades, but heavy metals already in the soil may be mobilized by local and global changes in soil conditions and exert toxic effects on soil microorganisms. We examined in vitro effects of Cd, Pb, and Zn on critical life stages in metal-sensitive ecotypes of arbuscular mycorrhizal (AM) fungi, including spore germin...

Influence of Glomus intraradices and Soil Phosphorus on Meloidogyne incognita Infecting Cucumis melo.

The interaction among Glomus intraradices, Meloidogyne incognita, and cantaloupe was studied at three soil phosphorus (P) levels in a greenhouse. All plants grew poorly in soil not amended with P, regardless of mycorrhizal or nematode status. In soil amended with 50 mug P /g soil, M. incognita suppressed the growth of nonmycorrhizal plants by 84%. In contrast, growth of mycorrhizal plants inocu...

Genetic diversity of the arbuscular mycorrhizal fungus Glomus intraradices as determined by mitochondrial large subunit rRNA gene sequences is considerably higher than previously expected.

Glomus intraradices is a widespread arbuscular mycorrhizal fungus (AMF), which has been found in an extremely broad range of habitats, indicating a high tolerance for environmental factors and a generalist life history strategy. Despite this ecological versatility, not much is known about the genetic diversity of this fungal species across different habitats or over large geographic scales. A n...

The Pythium suppressive ability of Glomus intraradices in cherry tomato propagation

This experiment aims at establishing the ability of the endomycorrhizal fungus Glomus intraradices to suppress the plant pathogen Pythium ultimum in cherry tomato. A study published in Canadian Journal of Plant Pathology in 1994 showed that Glomus intraradices suppresses Pythium ultimum in Tagetes patula (french marigold) (St-Arnaud, Fortin, Caron, & Hamel, 1994). Pythium ultimum and other Pyth...

Density dependence and interspecific interactions between arbuscular mycorrhizal fungi mediated plant growth, glomalin production, and sporulation

Functional differences between the arbuscular mycorrhizal fungi Glomus intraradices Schenk and Smith and Scutellospora heterogama Nicolson and Gerdemann as they affect Persea americana Mill. growth, glomalin, and fungal sporulation were examined by varying the composition and relative density of the two fungi over a gradient of available phosphorus (P). The plant benefit provided by these mycor...
